Solid-Phase Peptide Synthesis of ACP (65-74) Using PeptiPilot: Enhancing Peptide Synthesis Efficiency Guided by Process Analytical Tools
This application note demonstrates how integrated process analytical tools (PAT) within the PeptiPilot, a flow-based peptide synthesizer enables rapid method development and optimization for solid-phase peptide synthesis (SPPS). ACP (65-74) was selected as a model peptide to investigate the relationship between reaction temperature, synthesis time, and crude peptide quality under different synthesis conditions.
Real-Time Monitoring of Fmoc Deprotection Using PeptiPilot®

This application note demonstrates how PeptiPilot’s integrated UV detection system and PeptiSoft® software enable real-time monitoring of Fmoc deprotection and provide output on deprotection completeness and synthesis performance. Specialized instrument and software features, such as watches based on conditional UV absorbance thresholds and inclusion of fl ow interruption periods [2] facilitate complete deprotection in di icult to synthesize peptides.
